Показання до застосування β-інтерферонів людини в клінічній практиці

Препарати β-інтерферону людини — високоефективні імунотерапевтичні агенти з плейотропними біологічними та мультимодальними терапевтичними ефектами. Завдяки імуномодулювальній, протизапальній та противірусній дії β-інтерферонам відведено важливе місце в лікуванні рецидивно-ремітивного розсіяного склерозу у людей. У неврології в рамках персоніфікованої медицини ці імунотерапевтичні агенти можна використовувати також при автоімунних ураженнях периферичної нервової системи, деяких тяжких вірусних нейроінфекціях, енцефаліті Расмуссена, хворобі Альцгеймера і злоякісних новоутвореннях головного мозку. Серед екстрацеребральних показань до застосування β-інтерферонів слід виділити клітинні імунодефіцити, вірусні інфекції, зокрема папіломатоз і герпесвірусні ураження, цироз печінки, злоякісні пухлини, неспецифічний виразковий коліт, ревматоїдний артрит, автоімунний увеїт, респіраторний дистрес-синдром, кардіоміопатії, а також тромбоцитопенію у хворих з онкологічною патологією та в інфікованих вірусними інфекціями пацієнтів.

Показания к применению β-интерферонов человека в клинической практике

Препараты β-интерферона человека — высокоэффективные иммунотерапевтические агенты с плейотропными биологическими и мультимодальными терапевтическими эффектами. Благодаря иммуномодулирующим, противовоспалительным и противовирусным воздействиям β-интерфероны заняли важное место в лечении рецидивирующе-ремиттирующего рассеянного склероза у людей. В неврологии в рамках персонифицированной медицины эти иммунотерапевтические агенты можно использовать также при аутоиммунных поражениях периферической нервной системы, некоторых тяжелых вирусных нейроинфекциях, энцефалите Расмуссена, болезни Альцгеймера и злокачественных новообразованиях головного мозга. Среди экстрацеребральных показаний к применению β-интерферонов следует выделить клеточные иммунодефициты, вирусные инфекции, в частности папилломатоз и герпесвирусные поражения, цирроз печени, злокачественные опухоли, неспецифический язвенный колит, ревматоидный артрит, аутоиммунный увеит, респираторный дистресс-синдром, кардиомиопатии, а также тромбоцитопению у больных с онкологической патологией и у и инфицированных вирусными инфекциями пациентов.

Прогнозування функціонального наслідку раннього відновного періоду мозкового півкульного ішемічного інсульту за рівнем нейронспецифічної енолази в сироватці крові

Мета — розробити критерії прогнозування функціонального наслідку раннього відновного періоду мозкового півкульного ішемічного інсульту (МПІІ) за рівнем у сироватці крові нейронспецифічної енолази (НСЕ).
Матеріали і методи. Проведено комплексне клініко-інструментально-лабораторне обстеження 72 хворих (середній вік — (57,4 ± 1,2) року) в ранній відновний період МПІІ з використанням шкали NIHSS (National Institute of Health Stroke Scale), індексу Бартел, модифікованої шкали Ренкіна (МШР) на 10, 30, 90 і 180-ту добу захворювання, а також комп’ютерної томографії головного мозку. Рівень НСЕ в сироватці крові визначали на 10-ту добу захворювання. Для розробки критеріїв прогнозування використано ROC-аналіз.
Результати. Встановлено, що хворі на МПІІ, у яких оцінка за МШР на 180-ту добу захворювання становила ≥ 3 бали, мали вищий рівень НСЕ у сироватці порівняно з пацієнтами з оцінкою за МШР < 3 балів (3,54 [2,29; 4,44] та 1,56 [0,59; 2,58] мкг/л відповідно, р < 0,05). Рівень НСЕ > 1,4 мкг/л на 10-ту добу МПІІ є предиктором значення ≥ 3 бали за МШР на 180-ту добу захворювання (чутливість — 90,0 %, специфічність — 45,2 %).
Висновки. Вміст нейронспецифічної енолази у сироватц крові на 10-ту добу МПІІ більш інформативний для прогнозування рівня інвалідизації за модифікованою шкалою Ренкіна на 180-ту добу захворювання (AUC = 0,74), ніж рівня функціональної незалежності за індексом Бартел (AUC = 0,64, р < 0,05).

Прогнозирование функционального исхода раннего восстановительного периода мозгового полушарного ишемического инсульта по уровню нейронспецифической энолазы в сыворотке крови

Цель — разработать критерии прогнозирования функционального исхода раннего восстановительного периода мозгового полушарного ишемического инсульта (МПИИ) на основании определения уровня в сыворотке крови нейронспецифической энолазы (НСЭ).
Материалы и методы. Проведено комплексное клинико-инструментально-лабораторное обследование 72 больных (средний возраст — (57,4 ± 1,2) года) в ранний восстановительный период МПИИ с использованием шкалы NIHSS (National Institute of Health Stroke Scale), индекса Бартел, модифицированной шкалы Рэнкина (МШР) на 10, 30, 90 и 180-е сутки заболевания, а также компьютерной томографии головного мозга. Уровень НСЭ в сыворотке крови определяли на 10-е сутки заболевания. Для разработки критериев прогнозирования использовали ROC-анализ.
Результаты. Установлено, что больные МПИИ, у которых оценка по МШР на 180-е сутки заболевания составляла ≥ 3 балла, имели более высокий уровень НСЭ в сыворотке по сравнению с пациентами со оценкой по МШР < 3 балла (3,54 [2,29; 4, 44] мкг/л против 1,56 [0,59; 2,58] мкг/л, р < 0,05). Уровень НСЭ > 1,4 мкг/л на 10-е сутки МПИИ является предиктором значения ≥ 3 балла по МШР на 180-е сутки заболевания (чувствительность — 90,0 %, специфичность — 45,2 %).
Выводы. Содержание в сыворотке крови нейронспецифической энолазы на 10-е сутки МПИИ является более информативным для прогнозирования уровня инвалидизации по модифицированной шкале Рэнкина на 180-е сутки заболевания (AUC = 0,74), чем уровня функциональной независимости по индексу Бартел (AUC = 0,64, р < 0, 05).

Коморбідність мозкового інсульту за результатами прагматичного спостереження у м. Києві

Мета — вивчити коморбідність та чинники ризику виникнення мозкового інсульту в популяції хворих м. Києва.
Матеріали і методи. Проведено обстеження 1575 хворих  з гострим мозковим інсультом (з них 42,6 % чоловіків) віком від 26 до 100 років (середній вік — (70,5 ± 10,7) року), які були госпіталізовані в період з 1 червня до 31 грудня 2016 р. до неврологічних відділень м. Києва. Коморбідність вивчали із використанням індексу коморбідності Чарлсон.
Результати. Встановлено, що у 1102 (71,5 %) обстежених пацієнтів інсульт виник вперше. Переважали пацієнти віком 60 років і старші (84,1 %). Коморбідну патологію виявлено у 97,7 % пацієнтів. Діапазон значень за індексом Чарлсон — від 1 до 8 балів. Пацієнти з мозковим інсультом, який виник уперше, мали нижче значення індексу Чарлсон (у середньому — (3,5 ± 1,5) бала) порівняно з пацієнтами, котрі перенесли повторний мозковий інсульт ((3,8 ± 1,4) бала, р < 0,01). Значущо вищі значення індексу Чарлсон мали хворі з ішемічним інсультом порівняно з хворими з геморагічним інсультом (р = 0,012). Групи хворих із високим і низьким рівнем коморбідності за індексом Чарлсон статистично значущо відрізнялися за оцінкою за шкалою ком Глазго (p < 0,01), тяжкістю неврологічного дефіциту за шкалою NIHSS (National Institute of Health Stroke Scale) (p < 0,01) і тривалістю лікування в умовах неврологічного стаціонару (р = 0,38). Лише 49 % хворих із оцінкою за індексом Чарлсон ≥ 2 бали мали сприятливий наслідок за модифікованою шкалою Ренкіна в гострий період інсульту порівняно із 83 % пацієнтів, у яких індекс Чарлсон становив ≤ 1 бал (р < 0,01). Ризик летального наслідку в гострий період інсульту був пов’язаний з рівнем коморбідності за індексом Чарлсон (р < 0,01).
Висновки. Коморбідність, оцінена за індексом Чарлсон, пов’язана з тяжким перебігом та несприятливими наслідками гострого періоду мозкового інсульту. З огляду на велику поширеність серцевої патології у хворих з мозковим інсультом, більшу увагу слід приділяти профілактичним заходам, зокрема лікуванню первинного ураження серцево-судинної системи, та просвітницькій роботі серед населення.

Коморбидность мозгового инсульта по результатам прагматического наблюдения в г. Киеве

Цель — изучить коморбидность и факторы риска возникновения мозговых инсультов в популяции больных г. Киева.
Материалы и методы. Обследованы 1575 пациентов с острым мозговым инсультом (з них 42,6 % мужчин) в возрасте от 26 до 100 лет (средний возраст — (70,5 ± 10,7) года), которые были госпитализированы в период с 1 июня по 31 декабря 2016 г. в неврологические отделения г. Киева. Коморбидность изучали с использованием индекса Чарлсон.
Результаты. Установлено, что у 1102 (71,5 %) обследованных пациентов инсульт развился впервые. Возраст большинства пациентов составил 60 лет и старше (84,1 %). Коморбидная патология имела место у 97,7 % пациентов. Диапазон значений индекса Чарлсон — от 1 до 8 баллов. У пациентов с мозговым инсультом, возникшим впервые, значения индекса Чарлсон были значимо ниже (в среднем — (3,5 ± 1,5) балла) по сравнению с пациентами, перенесшими повторный мозговой инсульт ((3,8 ± 1,4) балла, р < 0,01). Значимо более высокие значения индекса Чарлсон были у пациентов с ишемическим инсультом по сравнению с пациентами с геморрагическим инсультом (р = 0,012). Группы пациентов с высоким и низким уровнями коморбидности статистически значимо отличались по оценке по шкале ком Глазго (p < 0,01), тяжести неврологического дефицита по шкале NIHSS (National Institute of Health Stroke Scale) (p < 0,01) и длительности лечения в условиях неврологического стационара (р = 0,38). Лишь у 49 % пациентов с индексом Чарлсон ≥ 2 балла отмечен благоприятный функциональный исход в острый период инсульта по сравнению с 83 % пациентов, у которых величина индекса Чарлсон составила ≤ 1 балл (р < 0,01). Риск летального исхода в острый период инсульта был связан с уровнем коморбидности по индексу Чарлсон (р < 0,01).
Выводы. Коморбидность, оцененная по индексу Чарлсон, связана с тяжелым течением и неблагоприятным исходом в острый период мозгового инсульта. Учитывая высокую распространенность сердечной патологии у больных с мозговым инсультом, следует уделять больше внимания профилактическим мерам, в частности лечению первичного поражения сердечно-сосудистой системы, и просветительной работе среди населения.

Прогностична оцінка розвитку когнітивних та емоційних порушень у хворих на розсіяний склероз залежно від локалізації вогнищ демієлінізації в головному мозку

Мета — дати прогностичну оцінку розвитку когнітивних та емоційних порушень у хворих на розсіяний склероз (РС) залежно від локалізації вогнищ демієлінізації в головному мозку.
Матеріали і методи. У дослідження було залучено хворих на РС з когнітивними розладами (n = 87), симптомом втоми (n = 92) та депресією (n = 79). Усім пацієнтам проводили магнітно-резонансне дослідження головного мозку на МР-томографі з індукцією магнітного поля 1,5 Тл. Визначення когнітивного дефіциту здійснювали за допомогою Монреальської шкали оцінки когнітивних функцій (MoCA). Для виявлення втоми використовували шкалу тяжкості втоми (Fatigue Severity Scale (FSS)). Депресію визначали за допомогою шкали депресії Бека. Обробку результатів проводили за допомогою методів описової статистики; для оцінки статистичної значущості показників використовували параметричні (t-критерій Стьюдента) та непараметричні (Т-критерій Вілкоксона) критерії. Для виявлення прогностичного значення отриманих даних обчислювали відношення шансів (ВШ).
Результати. Наявність атрофії мозолистого тіла статистично значущо підвищувала ризик розвитку когнітивних порушень у хворих на РС (ВШ 26 (3,40 — 198,9), р = 0,0001). Прогностична оцінка розвитку симптому втоми статистично значущо залежала від наявності атрофії мозолистого тіла (ВШ 2,82 (0,98 — 8,11), р = 0,047), вогнищ у тім’яних частках (ВШ 2,46 (1,02 — 5,9), р = 0,041) і зливних вогнищ (ВШ 2,58 (0,96 — 6,97), р = 0,05), розвитку депресії — від наявності атрофії мозолистого тіла (ВШ 2,81 (1,15 — 6,86), р = 0,021), вогнищ у скроневих частках (ВШ 5,22 (2,29 — 11,89), р = 0,001) та зливних вогнищ (ВШ 3,45 (1,42 — 8,41), р = 0,005).
Висновки. Локалізація вогнищ у головному мозку має прогностичну цінність щодо розвитку когнітивних та емоційних симптомів у хворих на РС.

Прогностическая оценка развития когнитивных и эмоциональных нарушений у больных рассеянным склерозом в зависимости от локализации очагов демиелинизации в головном мозге

Цель — дать прогностическую оценку развития когнитивных и эмоциональных нарушений у больных рассеянным склерозом (РС) в зависимости от локализации очагов демиелинизации в головном мозге.
Материалы и методы. В исследование были включены больные РС с когнитивными расстройствами (n = 87), симптомом усталости (n = 92) и депрессией (n = 79). Всем больным проводили магнитно-резонансное исследование головного мозга на МР-томографе с индукцией магнитного поля 1,5 Тл. Определение когнитивного дефицита осуществляли с помощью Монреальской шкалы оценки когнитивных функций (MoCA). Для выявления усталости использовали шкалу тяжести усталости (Fatigue Severity Scale (FSS)). Депрессию определяли с помощью шкалы депрессии Бека. Обработку результатов проводили с помощью методов описательной статистики; для оценки статистической значимости показателей использовали параметрические (t-критерий Стьюдента) и непараметрические (Т-критерий Вилкоксона) критерии. Для установления прогностического значения полученных данных определяли показатель отношения шансов (ОШ).
Результаты. Наличие атрофии мозолистого тела статистически значимо повышало риск развития когнитивных нарушений у больных с РС (OШ 26 (3,40 — 198,9), р = 0,0001). Прогностическая оценка развития симптома усталости статистически значимо зависела от наличия атрофии мозолистого тела (OШ 2,82 (0,98 — 8,11), р = 0,047), очагов в теменных долях (OШ 2,46 (1,02 — 5,9), р = 0,041) и сливных очагов (OШ 2,58 (0,96 — 6,97), р = 0,05), развития депрессии — от наличия атрофии мозолистого тела (OШ 2,81 (1,15 — 6,86), р = 0,021), очагов в височных долях (OШ 5,22 (2,29 — 11,89), р = 0,001) и сливных очагов (OШ 3,45 (1,42 — 8,41), р = 0,005).
Выводы. Локализация очагов в головном мозге имеет прогностическую ценность относительно развития когнитивных и эмоциональных симптомов у больных РС.

Вплив комплексної терапії з використанням мікрополяризації на електроенцефалографічні показники у дітей, хворих на симптоматичну епілепсію

Мета — проаналізувати можливість застосування методу транскраніальної мікрополяризації для поліпшення електроенцефалографічних показників у дітей, хворих на симптоматичну епілепсію.
Матеріали і методи. Обстежено та комплексно проліковано 29 хворих віком від 2 до 12 років з різними формами епілепсії. До групи порівняння, в якій було застосовано базисні лікувально-реабілітаційні заходи, залучено 14 дітей, до основної групи, в якій додатково проводили курс транскраніальної мікрополяризації, — 15. Транскраніальну мікрополяризацію виконували згідно з розробленими індивідуальними схемами лікування. За допомогою електроенцефалографії оцінювали показники біоелектричної активності головного мозку до початку лікування та через 3 міс після закінчення терапевтичного курсу.
Результати. Аналіз електроенцефалограм хворих дітей обох груп до лікування свідчив про наявність змін епілептичного типу (повільні множинні спайки, гострі хвилі, піки та епілептиформні комплекси). Після закінчення курсу транскраніальної мікрополяризації відзначено виражене поліпшення електроенцефалографічної картини (зменшення вираженості епілептиформних змін, збільшення частоти та амплітуди α- і β-ритмів, зменшення амплітуди та частоти δ- і θ-ритмів).
Висновки. Отримані дані свідчать про те, що використання методу мікрополяризації додатково до комплексного лікування хворих на епілепсію сприяє поліпшенню електроенцефалографічної картини і, відповідно, позитивно впливає на клінічний перебіг захворювання.

Влияние комплексной терапии с использованием микрополяризации на электроэнцефалографические показатели у детей, больных симптоматической эпилепсией

Цель — проанализировать возможность применения метода транскраниальной микрополяризации для улучшения электроэнцефалографических показателей у детей, больных симптоматической эпилепсией.
Материалы и методы. Обследовано и комплексно пролечено 29 больных в возрасте от 2 до 12 лет с разными формами эпилепсии. В группу сравнения, в которой проводили базисные лечебно-реабилитационные мероприятия, вошли 14 детей, в основную группу, в которой дополнительно проводили курс транскраниальной микрополяризации, — 15. Транскраниальную микрополяризацию выполняли в соответствии с разработанными индивидуальными схемами лечения. С помощью электроэнцефалографии оценивали показатели биоэлектрической активности головного мозга до начала лечения и через 3 мес после окончания курса терапии.
Результаты. Анализ электроэнцефалограмм больных детей обеих групп до лечения свидетельствовал о наличии изменений эпилептического типа (медленные множественные спайки, острые волны, пики и эпилептиформные комплексы). После окончания курса транскраниальной микрополяризации отмечено выраженное улучшение электроэнцефалографической картины (уменьшение выраженности эпилептиформных изменений, увеличение частоты и амплитуды α- и β-ритмов, уменьшение амплитуды и частоты δ- и θ-ритмов.
Выводы. Полученные данные свидетельствуют о том, что использование метода микрополяризации дополнительно к комплексному лечению больных эпилепсией способствует улучшению электроэнцефалографической картины и, соответственно, положительно влияет на клиническое течение заболевания.

Порівняльний аналіз динаміки рівня рухової функції паретичної кінцівки щура на тлі травми спинного мозку та відновних нейроінженерних втручань

Мета — порівняти динаміку рівня рухової функції на тлі однобічного перетину спинного мозку зрілого щура, трансплантації тканини нюхової цибулини (ТТНЦ) та імплантації макропористого гідрогелю в поєднанні з нейрогенними стовбуровими клітинами (НСК).
Матеріали і методи. Як первинні цифрові дані використано емпіричний матеріал, отриманий у попередніх дослідженнях (В. І. Цимбалюк та співавт., 2016, 2017). Тварини: зрілі (вік — 5,5 міс, маса тіла — 300 г) білі безпородні щури-самці (інбредна лінія, похідна Wistar) та щури-самці лінії Wistar. Експериментальні групи: «ЛПП» — лівобічний половинний перетин (ЛПП) спинного мозку на рівні Т11 (n = 40), «ЛППJUV» — ЛПП у тварини молодого віку (1 міс, n = 32), «ТТНЦ» — ЛПП + негайна гомотопічна ТТНЦ (n = 34), «NG» (NeuroGel) — ЛПП + негайна гомотопічна імплантація фрагмента макропористого гідрогелю (n = 20), «NG + НСК» — ЛПП + негайна гомотопічна імплантація фрагмента макропористого гідрогелю, асоційованого з НСК фетального (Е17) гіпокампа миші (n = 20). Моніторинг показника функції (ПФ) паретичної кінцівки проводили з використанням шкали Вasso — Вeattie — Вresnahan, розрахунок щотижневого приросту ПФ, прискорення приросту, а також статистичний аналіз — із застосуванням програмного пакета Statistica 10.0.
Результати. Максимуми щотижневого приросту ПФ у групах «ЛПП», «ЛППJUV» і «ТТНЦ» відзначали на 1-му та 2-му тижні, у групі «NG» — на 1, 2, 3, 6 і 7-му тижні, у групі «NG+НСК» — на 1, 2, 3, 6, 7, 8 і 12-му тижні, максимуми щотижневого прискорення приросту ПФ — на 1-му («ЛПП», «ТТНЦ»), 1-му і 5-му («ЛППJUV»), 1-му та 6-му («NG + НСК»), 1, 2 і 6-му («NG») тижні спостереження.
Висновки. Враховуючи отримані дані та фазність перебігу спінальної травми, можна припустити, що ТТНЦ обмежує ранні реакції вторинної альтерації, імплантація макропористого гідрогелю більшою мірою стимулює ранні нейропластичні реакції та ремієлінізацію волокон перифокальної зони, трансплантація НСК у комплексі з макропористим гідрогелем, окрім зазначених механізмів, стимулює пластичність нейрональних мереж і ріст нервових волокон крізь зону ураження у проміжний та пізній період травми.

Сравнительный анализ динамики уровня двигательной функции паретической конечности крысы на фоне травмы спинного мозга и восстановительных нейроинженерных вмешательств

Цель — сравнить динамику уровня двигательной функции на фоне одностороннего пересечения спинного мозга зрелой крысы, трансплантации ткани обонятельной луковицы (ТТОЛ) и имплантации макропористого гидрогеля в сочетании с нейрогенными стволовыми клетками (НСК).
Материалы и методы. В качестве первичных цифровых данных использовали эмпирический материал, полученный в предыдущих исследованиях (В. И. Цимбалюк и др. 2016, 2017). Животные: зрелые (возраст — 5,5 мес, масса тела — 300 г) белые беспородные крысы-самцы (инбредная линия, производная Wistar) и крысы-самцы линии Wistar. Экспериментальные группы: «ЛПП» — левостороннее половинное пересечение (ЛПП) спинного мозга на уровне Т11 (n = 40), «ЛППJUV» — ЛПП у животных молодого возраста (1 мес, n = 32), «ТТНЦ» — ЛПП + немедленная гомотопическая ТТОЛ (n = 34), «NG» (NeuroGel) — ЛПП + немедленная гомотопическая имплантация фрагмента макропористого гидрогеля (n = 20), «NG + НСК» — ЛПП + немедленная гомотопическая имплантация фрагмента макропористого гидрогеля, ассоциированного с НСК фетального (Е17) гиппокампа мыши (n = 20). Мониторинг показателя функции (ПФ) паретической конечности проводили с использованием шкалы Вasso — Вeattie — Вresnahan, расчет еженедельного прироста ПФ, ускорения прироста, а также статистический анализ — с применением программного пакета Statistica 10.0.
Результаты. Наибольшие значения еженедельного прироста ПФ в группах «ЛПП», «ЛППJUV» и «ТТОЛ» отмечали на 1-й и 2-й неделе, в группе «NG» — на 1, 2, 3, 6- и 7-й неделе, в группе «NG + НСК» — на 1, 2, 3, 6, 7, 8 и 12-й неделе, наибольшие значения еженедельного ускорения прироста ПФ — на 1-й («ЛПП», «ТТОЛ»), 1-й и 5-й («ЛППJUV»), 1-й и 6-й («NG + НСК»), 1, 2 и 6-й («NG») неделе наблюдения.
Выводы. Учитывая полученные результаты и фазность течения спинальной травмы, можно предположить, что ТТОЛ ограничивает ранние реакции вторичной альтерации, имплантация макропористого гидрогеля в большей степени стимулирует ранние нейропластические реакции и ремиелинизацию волокон перифокальной зоны, трансплантация НСК в комплексе с макропористым гидрогелем, кроме указанных механизмов, стимулирует пластичность нейрональных сетей и рост нервных волокон через зону повреждения в промежуточный и поздний период травмы.

Ефективність використання зеленого флуоресцентного білка як маркера для ідентифікації трансплантованих нейральних прогеніторних клітин у нервовій тканині мишей

Мета — оцінити ефективність використання зеленого флуоресцентного білка (GFP) як маркера донорських клітин у тканинах реципієнта у віддалені строки після трансплантації.
Матеріали і методи. Експериментальним тваринам трансплантували нейральні прогеніторні клітини, виділені з гіпокампів мишей лінії FVB-Cg-Tg(GFPU)5Nagy/J, трансгенних за геном GFP. Через 90 днів після трансплантації GFP-позитивних нейральних прогеніторних клітин проведено імуногістохімічний аналіз зрізів мозку із використанням антитіл проти GFP за допомогою конфокального та електронного мікроскопа.
Результати. Імуногістохімічне дослідження з використанням антитіл проти GFP показало, що на 90-ту добу після стереотаксичної трансплантації у гіпокамп GFP-позитивні клітини диференціювалися в клітини з нейронною та гліальною морфологією. Ультраструктурний аналіз виявив, що трансплантовані GFP-позитивні клітини утворювали синаптичні контакти між донорськими клітинами і нейронами реципієнта на 90-ту добу після трансплантації.
Висновки. Результати дослідження свідчать про те, що GFP — це надійний нетоксичний маркер, який дає змогу ідентифікувати донорські клітини навіть у віддалені (до 90 діб) строки після трансплантації. Імуногістохімічне фарбування зрізів мозку із використанням антитіл проти GFP дає змогу візуалізувати донорські GFP-позитивні клітини за допомогою як конфокального, так і електронного мікроскопа.

Цель — оценить эффективность использования зеленого флуоресцентного белка (GFP) как маркера донорских клеток в тканях реципиента в отдаленные сроки после трансплантации.
Материалы и методы. Экспериментальным животным трансплантировали нейральные прогениторные клетки, которые были выделены из гиппокампов мышей линии FVB-Cg-Tg(GFPU)5Nagy/J, трансгенных по гену GFP. Через 90 дней после трансплантации GFP-положительных нейральных прогениторных клеток был проведен иммуногистохимический анализ срезов мозга с использованием антител против GFP с помощью светового и электронного микроскопа.
Результаты. Иммуногистохимическое исследование с использованием антител против GFP показало, что на 90-е сутки после стереотаксической трансплантации в гиппокамп GFP-положительные клетки дифференцировались в клетки с нейронной и глиальной морфологией. Ультраструктурный анализ выявил, что трансплантированные GFP-положительные клетки образовали синаптические контакты между донорскими клетками и нейронами реципиента на 90-е сутки после трансплантации.
Выводы. Результаты исследования свидетельствуют о том, что GFP — это надежный нетоксичный маркер, позволяющий идентифицировать донорские клетки даже в отдаленные (до 90 суток) сроки после трансплантации. Иммуногистохимическое окрашивание срезов мозга с использованием антител против GFP позволяет визуализировать донорские GFP-положительные клетки при помощи как светового, так и электронного микроскопа.
